Looking at trichomes

I purchased a 60x-100x microscope and set about trying to see the trichomes. The problem is (and no one has explained this fully) what part do you pick to view them. If you take a small leaf from the bud and view that, will that give the same picture as the rest of the bud?

It's hard to look at them without taking a bit of the plant off and looking on a flat surface such as a table.

So my question is... Can I take a leaf sample and use that as a guide to harvest?

Thanks. I take it I am looking for 50/50 cloudy and amber.

Out of interest, do many people do sampling (cutting, quick drying and smoking) to determine when to harvest?
