Help with Grow Room

Currently I've got some babies in a room underneath the stairs (most people have a bathroom here). It's not that large and I'm not there currently so I don't know the dimensions. It has one vent going to this room that I haven't felt air coming out of so it could be a return.

I have a fan blowing in the room right now and the door open to help with the temperature from a 1000w MH light. I need to know what to do for ventillation in this room or if I should just move it to another room.

My current plan is to put an in-line fan and some ducting onto the light to help cool it and to exhaust some hot air through the vent. If I do it this way, will there be enough passive air from underneath the door?

Yep you ought to use a large in-line vent fan to cool the light and also room and I'd add a charcoal filter in that line too. Then try to vent it outside somehow. (through the vent you mentioned? IF it was meant to be a bathroom perhaps that was the exhaust vent for it?) There will probably be enough air leak under and around the door to replace the vented air. Otherwise it'll have to suck it from a hole you make. My advice on the fan is to go big.

IF you own the house and can cut holes, you can make it work really easy.

Having that return vent makes it nice too.
