Need some help with my grow setup.

So I'm going to be growing in a room that is about 15'x10'x12' approx. There is rather large (2x2 feet) a/c duct in the middle of the ceiling that will be constantly blowing cool air in the summer, and hot air in the winter, with a another exhaust vent about the same size at the back end of the room.

I'm going to be growing in a completely enclosed space built by me within the room made from a simple wood frame and lined with black/white plastic. Inside it will be divided between flower and veg rooms.

As far as the actual room goes, the walls and floor are carpeted, and the ceiling is basically old nasty looking ceiling tile covered in dust and above that, god knows what.

This is my first grow so i am a complete novice.

In the flower room I will be running two 1000w lights. The veg will have a simple floro set up for the clones and a 250w MH light for the veggies.

Inside the grow enclosure the plastic walls will be lined with mylar and/or cheap fire blankets.

And now to the questions.....

1. There are no windows, just a single door leading into the room, which will be locked AT ALL TIMES. This leaves me with only one source of outside air, the a/c duct. Will this be fine?

2. There is dust EVERYWHERE on the ceiling tiles, a/c and exhaust vents, and even up on high to reach places in the room. I can clean the vent grilles, but as far as reaching up their ducting, its a lost cause, and there's no telling whats up in there. In order to circulate air, if i leave the enclosure door flaps open for an extended period of time, say no more than a few hours a day (2-3), would they be at all susceptible to diseases?

3. Can I run my vent fan ducts together along with my regular exhaust duct that sucks the air out of my veg and flower rooms and have it all shoot right out the exhaust vent in the room?

I'm sure ill have way more questions up to and including after i start building this setup.
