Building a room with darkroom vents.

So I building a room a a I have some dark room vent I have installed. Its about a 5 x5 I have a six inch fan I think like 400 ish cfm carbon filter all that good stuff. Few questions... are the two vents for intake going to be enough given they are dark room vents and might restrict air flow? I believe they are 8x8 square. Also in building the wall door frame area I am wondering do I need to caulk the inside of my walls on one side before installing my installation and the other side of the wall? I'm not going for a totally sealed room but I dont want light leaks.
