11-13% moisture content is a good range for bucking off the stem. We typically start the cure process, and after about 3 weeks curing is when we'll bring samples in to test. This is mainly to get the better terpene profile. Is this a personal grow or commercial?

.... sorry, and as far as measuring the moisture content, I use a wood moisture probe I got off of AMZ like this one.....

If you're in a legal State, you can get a Potency and Terps test done for like $100. If you don't care about terps (which you should), Potency alone is like $50. At least here if you are not Commercial, I think all you have to have is your Patient Card.
edit: Sample size is going to be about 5g.

I've read somewhere that you need to completely dry the bud before using it. It takes around 5-7 days for the buds to fully dry, however it will vary depending on the climate and drying location. Hope this help!
