I've actually thought about this subject a lot... it's suicide of course, unless you get picked up hitchhiking somewhere in the galaxy...
I think it would be great to have a vaporizer, enough food, water and cannabis for a year, my MP3 player and the views of space all around me in a little cocoon.
Just remember to hang around in LEO and build up that escape velocity (10.9km/s) or else you'll go nowhere.

You guys are thinking about it wrong...
Were not bringing stuff- we need to bring the means to produce things and the people who know how to do it and how to build repair and maintain equipment- Think of it as a mobile planet- we need to produce everything on board to sustain generations, it's not about trying to pack enough stuff.

You'd have to build this ship in space you know... like a space dry-dock.
It'd be too heavy to lift off from earth with everything you're thinking of with current propulsion methods.
The moon would be perfect for building large space ships, except for all the collisions with asteroids the moon takes.
